mmCSM-AB: in silico approach for guiding antibody design

mmCSM-AB is a tool for analysing the effects of introducing multiple point mutations on antibody-antigen binding affinity. Users can predict the effects of a known mutation (Fast Mode), or systematic exploration of all potential mutations to guide rational antibody engineering (Design Mode).

The mmCSM-AB models were assessed on two experimentally characterised blind-test sets: based on (1) double and triple mutation constructs from SKEMPI2.0, mmCSM-AB achieved Pearson and Spearman correlations of up to 0.94 and 0.85 (RMSE of 1.12 Kcal/mol) (2) constructs with >3 mutations, mmCSM-AB achieved Pearson and Spearman correlations of up to 0.74 and 0.64 (RMSE of 1.56 Kcal/mol).
