Submission

1. Choose Organism of interest: FASTA sequences for all organisms are available for download, showing structure numbering used in the website.
2. Submit mutation: either as a single point mutation format or as a mutation list. Sample lists are available for download showing formatting required.

3. Single Point Mutation visualisation:
The ‘Results’ section shows the predicted outcome from our tool, and specific mutational details.
‘Interactive Viewer’ section enables the user to visualise the protein structure and respective interactions in a specific way: different molecules and interactions can be shown or hid. This viewer also enables molecule rotation, transposition, spinning and zoom, with added screenshot and PyMOL session downloads.

4. Mutation List visualisation:
The ‘Prediction’ section shows the list of mutations with their respective position details, and the predicted phenotype according to our tool.
The ‘details’ column permits the user to access the full details for each mutation submitted, in a similar format to point 3.
The ‘Interactive Viewer’ section visualises how all the mutations within the submitted list map across the RNA polymerase complex, where resistant mutations are shown in red, susceptible in blue, and any positions submitted having different phenotypes in green. This viewer also enables molecule rotation, transposition, spinning and zoom, with added screenshot and PyMOL session downloads.
